NK cell activity was first observed in human peripheral blood mononuclear cells (8, 9) and rodent splenocytes (5, 6); however, these large granular lymphocytes are known to reside in multiple lymphoid and non-lymphoid tissues including the bone marrow (BM), lymph nodes (LNs), skin, gut, tonsils, liver, and lungs . Natural killer (NK) cells can swiftly kill multiple adjacent cells if these show surface markers associated with oncogenic transformation. We further summarize the abundant preclinical attempts at overcoming these challenges, present past and ongoing clinical trial data and finally discuss the potential impact of novel insights on the development of NK cell-based therapies. NK cell cancer immunotherapy is an emerging anti-tumour therapeutic strategy that explores NK cell stimulation. ▪ Abstract NK cells are regulated by opposing signals from receptors that activate and inhibit effector function. In this article, we review our current knowledge of NK cells' biology, and highlight the challenges facing NK cell antitumor strategies in solid tumors. While positive stimulation may be initiated by an array of co-stimulatory receptors, specificity is provided by inhibitory signals transduced by receptors for MHC class I.
